THURSDAY, Jan. 24 (HealthDay News) -- A drug that nearly doubles the life span of mice with inherited amyotrophic lateral sclerosis (ALS) has been identified by University of Iowa researchers, who made the finding after discovering an unexpected reaction between proteins in the lab.

The study results, published online Jan. 24 inThe Journal of Clinical Investigation, may lead to treatments for some forms of ALS (also known as Lou Gehrig's disease), a fatal neurodegenerative disease that destroys motor nerve cells in the brain and spinal cord.
1월 24일판 클리니칼 인베스티게이션의 온라인 저널에 발표된 실험 결과에 의하면 어느 종류의 ALS(루게릭이라고도 알려진)를 치료할 수 있는 가능성을 제시하였다. ALS는 척수와 두뇌의 신경세포를 파괴하는 치명적인 퇴행성 신경질환이다.

The researchers were studying the basic biology of cell signaling when they made the unexpected discovery that superoxide dismutase-1 (SOD1) -- a protein that's mutated in inherited forms of ALS -- interacts with Rac1, a protein that regulates production of reaction oxygen species (ROS) by the Nox2 protein complex. ROS is essential for normal cell function, but abnormal ROS production is a suspected cause of ALS and other neurodegenerative diseases.
연구원들이 세포의 신호에 대한 기본적인 생물학을 연구할때 SOD1의 우연히 발견이 있었다. SOD1 단백질은 유전적인 성향을 가진 ALS의 면역력을 가졌다. SOD1은 Rac1이라는 Nox2라는 단백질 복합물로 의하여 활성산소종(ROS)을 통제하는 단백질과 상호작용을 한다. 활성산소종(ROS)는 근본적으로 정상적인 세포기능이지만 비정상적인 활성산소종(ROS)의 생산은 ALS나 다른 근위축성병의 원인으로 의심된다.

This unexpected interaction prompted the researchers to take a closer look. They first found that deletion of the Nox2 protein almost doubled the life span of mice with inherited ALS. This provided further evidence that Nox-2 generated ROS may play a role in ALS progression. The researchers then found that a drug called apocynin, which blocks Nox2, slows ALS progression and increases life span of mice with inherited ALS.

About 5 percent to 10 percent of ALS cases in the United States are inherited, while 90 percent to 95 percent of ALS cases are not inherited and can affect anyone.
미국내 ALS환자들중 대략 5~10%의 환자들이 유전적 성향을 가진 ALS를 가지고 있다. 나머지 90~95%의 환자들은 후천적인 성향을 가지고 있다.

There are no mouse models for sporadic ALS, so the researchers were unable to test whether apocynin has any effect on sporadic ALS. The researchers also noted that extensive safety and efficacy testing in pre-clinical trials must be conducted to determine if apocynin is effective in people.
이 연구소에선 후천적 성향을 가진 쥐를 확보하지 못하여 아포사이닌(apocynin)이 후천적 ALS환자에게 영향력이 있는지에 대해서는 아직 연구하지 못하였다. 연구원들은 아포사이닌(apocynin)이 다른 환자들에게도 효과가 있는지에 대한 포괄적인 안정성과 효율성이 함유된 연구가 진행되어야 된다고 말했다.

The discovery of the interaction between SOD1 and Rac1 may prove to have widespread import!!ance.

"It is exciting not only from the standpoint of ALS, but also because of its implications for understanding basic cell biology. Rac1 is implicated in many cellular processes including cellular migration, proliferation and differentiation, and is an import!!ant component of inflammatory disease processes," team leader John Engelhardt, head of anatomy and cell biology at UI's Roy J. and Lucille A. Carver College of Medicine, said in a prepared statement.
